是德国汉堡化学兼细菌学家(Gustav Giemsa，1867～1948)首先调制应用于生物染色剂之一。固体为暗红色或褐色粉末状，内主要含有二号天青(azureⅡ)和二号天青-曙红(azure Ⅱ-eosin)的混合物。应用时多溶于等量的甘油和甲醇中。当今市售已配好的暗红色吉氏溶液由天青B:二号天青-曙红:(碱性)亚甲蓝二1:12:2(质量比)组成，然后溶于甘油：甲醇=5:24(体积比)中，总浓度为0.6％(质量)，密度(20℃)0.89g/cm3。生物标本在其中染色时间约为45min。质量标准

Staining Procedures, 4th edition, Williams & Wilkins, 173 (1981); together with May-Grünwald Solution for pap
staining according to Pappenheim.
Principle:
When blood films are stained using Giemsa Stain, the nucleus and cytoplasm of white blood cells take on
characteristic blue or pink coloration. The use of purified eosin and thiazine dyes minimizes lot-to-lot variation.
Product Description:
Solution of azure B/azure II-eosin/methylene blue 1:12:2 (w/w/w) in glycerol/methanol 5:24 (v/v); total dye
content: 0.6 % (w/w)
Prior to use the Giemsa solution should be filtered and diluted (e.g. 1:20) with buffer solution pH 6.5 according
to Weise (Fluka 82632).
Fixation of slides:
Fix 10 min. in methanol; air-dry; immerse 45 min. in Giemsa Solution; rinse with distilled water; air-dry
